[Modern Research]
1. Chemical constituents: All parts of Datura contain alkaloids, with the highest content in the flowers. The main alkaloid is scopolamine, followed by atropine. The highly toxic components are scopolamine, atropine, and hyoscyamine.
2. Pharmacological effects: Flos Daturae has significant sedative effects. When combined with hypnotics such as nitrazepam or pentobarbital, it produces a strong synergistic effect. However, it has varying excitatory effects on the medulla and spinal cord, indicating a bidirectional effect of scopolamine on the central nervous system. It relaxes bronchial and gastrointestinal smooth muscles and inhibits bronchial mucus secretion; it relieves vagal inhibition on the heart, allowing sympathetic dominance and increased heart rate; it reduces gastrointestinal motility and tension, blocks cholinergic nerves, relaxes the detrusor muscle of the bladder, contracts the urethral sphincter, causing urinary retention; total alkaloids have antioxidant and free radical scavenging effects.
3. Clinical reports:
1) Chronic bronchitis: 15 g of Flos Daturae ground into fine powder, mixed with 500 ml of 60° grain alcohol, shaken and sealed for 7 days. Taken 3 times daily, 1–2 ml each time, maximum 2 ml daily. After one course (500 ml) if no cure, treatment can continue. Among 100 cases treated, 33 cured, 55 effective, 12 ineffective, total effective rate 88%. [Xinjiang Traditional Chinese Medicine, 1990,(1): Feng San]
2) Ankylosing spondylitis: Flos Daturae injection (2 ml per vial, each 1 ml equivalent to 0.5 ml scopolamine) or tincture administered intramuscularly before sleep or orally once daily. Among 34 cases, 21 markedly effective, 10 effective, 3 ineffective. [Journal of Traditional Chinese Medicine, 1988, 29(4):286]
3) Acute closed soft tissue injury: Treatment group (304 cases) and control group (101 cases) received Flos Daturae injury plaster and Musk Zhui Feng plaster respectively in a double-blind controlled trial. The treatment group had a 96.3% total clinical effective rate at 12 days, significantly better than control (P<0.05). [Jiangsu Traditional Chinese Medicine, 2000, 21(11):12]
4) Middle-aged and elderly Meniere’s disease vertigo: Treatment group applied Flos Daturae plaster behind the affected ear, changed every 3 days, 10 plasters per course; control group took oral Sibelium 10 mg daily before sleep. Each group observed 30 cases; total effective rates were 83.3% and 40.0% respectively, with a highly significant difference (P<0.005). [Chinese Journal of Integrated Traditional and Western Medicine in Otorhinolaryngology, 2001, 9(4):193]
